dc.description.abstract |
The aim of this study was to determine genetic diversity of Tripenustes gratilla (Linnaeus 1758) in Manokwari waters based on cytochrome oxidase I (COI) gene of the mitochondrial genome. We extracted DNA, amplified, cleaned and sequenced PCR products on 34 samples of T. gratilla. Genetic diversity analyses were performed using Mega5 dan DnaSP 5.10.01. We sequenced a 592-bp and the nucleotide composition average of this gene is T (34,42%), A (27,43%), C (22,76%) and G (15,38%), respectively. There are fifteen polymorphic sites and twelve haplotypes were identified in COI gene of T. gratilla of Manokwari. The haplotype diversity (Hd) and nucleotide diversity (π) values were 0.75 and 0.00248. We also identified 15 nucleotides in the synonymous substitution and they are in transition. No one nucleotide in transversion, insertion, and deletion. |
